Promega now offers broadest and fastest offerings for cell-free expression of proteins

New Insect Expression Version of TNT® Technology

MADISON, Wis.–(BUSINESS WIRE)–Promega Corporation announces it has expanded its offerings within its TNT® technology to include an insect expression system and an improved system for expression in bacteria. The recent additions give scientists the most comprehensive offering of cell-free expression tools allowing the expression of proteins from DNA in a single reaction.

The latest addition of an insect lysate system delivers unique advantages, most importantly expression at a lower temperature than other systems. Working in lower temperatures can enhance protein folding and stability and is often the preferred method for scientists performing mutagenesis studies.

The TNT system is a single step process that can be done in a fraction of the time of the other multi-stepped offerings in the market. The proprietary TNT technology from Promega utilizes DNA as template for the coupled transcription/translation, and saves researchers from purifying and quantifying mRNA for in vitro expression, greatly streamlining the process. “Expanding applications have placed demands on performance for protein expression in vitro in terms of yield and consistency,” says Gary Kobs, product manager for Promega’s cell-free expression systems. “Since its launch in 1992, the primary application for TNT has evolved from confirming protein expression to examining protein-protein interactions. As a tool for protein-protein interaction, we know scientists not only want greater yield, but also need to express proteins in a variety of systems such as insect cell extract.”

Cell-free protein expression offers unique advantages when compared to cell-based protein expression including time savings and increased overall yields of functional, soluble, full–length proteins. Typical reactions produce sufficient quantities of protein that can be used for a variety of applications including protein:protein and protein:nucleic acid interactions.
